I tried putting in all the villagers amigo card numbers in order but didn’t work

I don't think so, but iirc Wisps instrument(? the town tune that plays when u talk to someone) is spooky chimes or something.

Isabelle has a few Amiibo cards, her normal one is #001, but there's also #113 (Winter), #215 (Kimono), #301 (Dress), and #401 (Special Edition).

Nah, there isn't. Fauna's in 1 and Flora's in 3, for example.

I think it's refering to the Villager Trading Plaza. Flora, Fauna, Daisy, and Buck are all villagers.

I do wonder if the clue is referring to the New Leaf: Welcome Amiibo update specifically. I do think we're on the right track, though. I think it does have to be NL:WA or amiibo related.
