This has been a pressing question of mine when you tell a villager "Don't go!" And they stay is it possible for them to try and move again after that point

Yes. A villager can try to move out many times. The same villager will never attempt to move out twice in a row before another villager asks to move out.

No, they will at some point as to move again, unfortunately.
