So, having secured the Wii U Console from a fortune cookie, and having completed each of the 30 levels in Desert Island Escape with at least target score, I can say, with certainty, that it has been the most enjoyable aspect of the game for me.

Initially, I played only using Play Coins, but that got tedious, and frankly, I am impatient, so I ran out and bought an NFC reader (have the 2DS), and two packs of Series 2 Amiibo cards. Though I only got 9 unique cards out of 12, the cards I did get allowed me to build the perfect team for reaching my goals.

Ultimate team used:
Dobie - 4 Steps - 2 Food - Lone Wolf Ability
Buzz - 4 Steps - 1 Food - Hunter Ability
Sprocket - 6 Steps - 1 Food - Going the Distance Ability

Dobie with his Lone Wolf ability allowed me to send him in one direction, and the other two as a team in the other. Though Dobie would not collect items to build tools or medicine to heal, his not returning to camp with the others made exploring more efficient. Putting Buzz in front of Sprocket allowed Buzz to clear clouds while Sprocket came from behind to move the potential campsite farther.

With this team, I finally cleared the rest of my target scores, and ended up securing all the major prizes, including:

Desert Island Map - All target scores in Beginner
Desert Island Table - All target scores in Intermediate
Desert Island Stool - All target scores in Advanced
Ikada Trophy - All target scores, all maps

The ACNL Desert Island Wiki page will be updated to include the prizes for reaching target scores in Intermediate and Advanced.

If you have any questions or suggestions, add them here. I did feel a sense of fulfillment having beaten all levels with target score or higher.

This is really helpful information, thanks! I played a bit this morning and kept coming up about 2k shy of the scores needed, so I'll keep an eye out for characters with these abilities.
 
I just played the first level yesterday - it was fun! Is there any prize for completing the game?

Unfortunately, the only AC-compatible amiibo I have is Wolf Link, and he doesn't work on Desert Island Escape. I ended up having to shell out 6 play coins for the level. One of the characters I chose was Melba. Love the girl, but her special characteristic meant that I had to spin a roulette wheel and she'd feel too lazy half the time to even take her turn - ha!

How much did the amiibo cards cost you? I wouldn't mind picking a pack up, or maybe a preowned figurine at gamestop if the price is right.

You can get a pack of 6 on amazon for like $5. I only ever got 1 pack so I could play along with the expansion (you also get 5 MEOW coupons a day just by scanning a card).

I actually love this game! I have a bit of a love/hate relationship with the lone wolf types. I've been using Plucky/Cleo/Boris and I'm a pretty big fan of that team for now.
 
Thanks I will try that. I was stuck in intermediate with a Soft-Footed, Hearty traveler and First Aid.

Yes, you get some Wii U specific items depending on the difficulty of the level you beat, and I believe you get the Amiibo Game Shelf upon completion of all levels, regardless of score on each level.

And I paid $6 a pack at Target for mine.
