I just recently acquired an amiibo card for Hopper and I’m wondering exactly how it works. I have a villager that I want to replace with hopper by he’s my most recent move in. Does it matter?

So, with Amiibo visitors, you invite them 3x to your island before they move in. To invite them, go to your Resident Services -> the Nook machine. You should have an option to scan the amiibo then, and they'll show up in your campsite same day. They'll ask you to make them an item; if you don't have the DIY for the item, they'll give it to you, which can actually get some neat DIYs in the process. After the 3rd day and the 3rd gift, if you keep talking to them, you'll be able to ask them to move in. In their dialogue, they'll prompt you to select a current villager to kick out. It's fine if it's the most recent move-in, as long as they are all the way moved in (e.g., no longer in boxes / unpacking).
