Today, I loaded a new save file to check if all my villagers are still there. To my surprise Diana left! Is there a way I can get her back?
 
Nope, she's gone. You will need to cycle through 16 villagers to be able to have her in your town again.

So she was still there the last time you played, not in boxes? If so, TT back to that day and she'll still be there like no time has passed. Get her to ping you about moving and tell her to stay.
 
If you loaded a new save file and NOT your mayor or another existing character, then turn the 3DS SETTINGS CLOCK back until her house is still there. Do not use the game clock, that method won't work.

Unfortunately if you loaded your mayor or another character, then she's gone.

The game clock will work fine if they go directly back to the last day they played (I've done that before). If they can't remember the last day they played, then yeah, going back day by day with the 3DS clock would be safer.
 
I've found that even with a new save file, if the house has already disappeared, then it is gone no matter how you travel or with who. This is also true if she is in boxes... save file, TT, it doesn't matter. Boxes or the house gone is a bad sign.

I've also found that when plot setting with a new save file, if you get carried away with your TT (resetting several times is fine, I kept TTing out of frustration and impatience) and a new villager's house rather than fence is built, then the house will remain in that location. Traveling back made the fence reappear and even disappear, but the game has already established it, so it will still be there in the end. As long as it's only the fence you see every time, plot reset will work.

this isn't true. loads of people have started with a new save file to check if they've moved, they had, did NOT save file, and just changed back to the last date they played / to an earlier time (as the others have posted). then you just stop the villager because they are considering moving, obviously. as long as you haven't started up with your mayor.

i don't know about tt, but oftentimes when plot resetting, the house can stay stagnant in a certain location multiple times. that does not necessarily mean that it is stuck there forever. as long as you haven't saved with your new file, and as long as you hadn't accidentally started with your mayor, it will eventually move. the fence will only disappear if you have less than 8 villagers and it's a random move in because you aren't guaranteed a move in until, what, seven days i think?
 
As long as you did not save when you did the new save file trick, when you tt back to whatever is the last time you played and saved, the villager house should be there. I find that usually when you come back to real time or the last time you played, that villager will ping you. Good luck. :)

What I posted was my personal experience with it. It is possible that while I did TT backwards, I might not have went back to the last time I played causing it not to work for me. Who knows? I've just noticed that in boxes/no house = no good. I am experienced enough at TT so that villagers don't move out....so obviously, I do not have a lot of experience with what to do if they did. I am glad to have information that you can retrieve a lost villager. Too bad that most people start with their mayor because often, they don't know the villager left.

I am also very experienced at plot resetting, it was 100% due to TT when the problem came about. The fence will move as long as it is a fence, and it should stay that way if you are simply resetting and using the new save file...I used TT multiple times after appearance of the fence. That wasn't a good idea, so I made note of it. I agree the fence plot can become "stagnant" and will move eventually with resets and continuing with the new save file.

Careful, if you are going back day by day, you must keep using a New File every day to do that so that the game does not save. And once the house reappears, she'll be in boxes that day, so you need to go back even further so she has a chance to ping.
