You can purchase them on Animal Crossing: City Folk on Sundays. On Sunday Mornings (6am to 12pm) a lady named Sow Joan will be in your town. Talk to her and you can purchase the Turnips from her.

No. Tom Nook will randomize his buying price so you have a chance of profiting a lot or simply losing money =P

I'd buy more than 10,000 Turnips every time Joan was in my town and was selling them for an uber low price.

Remember that your best chance at good money is either friday or saturday morning... that when they are the best time to sell them (best priced those mornings)
