So say I have a villager moving out on their own will and I want to get an amiibo card. Can I put the land on "hold" or temporarily not sell it so I can save that space for a specific villager? That way another random villager won't take it.

I don't think that you can do this. However if that land does get taken, you can get your amiibo villager to kick them out

Okay then, given that the villager moving out just asked if they should move today, how many days until that land is sold to another random villager?

Don’t quote me but I believe it goes like this:
Day 1 - they ask to leave
Days 2 - the villager that’s moving will be in boxes
Day 3 - the plot is empty and will remain unsold for the day unless you invite a villager via island/amiibo/from someone else’s island
Day 4 - the plot will be sold randomly if you didn’t invite anyone on day 3

and there’s no way to delay or change this series of events at least that I’m aware of. However you can boot any villager via amiibo card at any time (after the 3 days of inviting said amiibo and completing their requests)

And to complement @Feferily answer, after the third day, you can select which villager you want them to replace, so don't worry if your plot is taken.

This is a nice idea, though! like since we cant remove plots let us at least put them on hold.
