I'm sure this has probably been asked thousands of times or at least mentioned here and there but I'm kind of lazy to do a search bar search for my answer. Does anyone know how animals moving in works in terms of location? What I mean is, is there any way to prevent them from moving in on top of your flower patches for example? I'd like to have my flowers and hybrids left in peace then to come back the next day to have someone plop their house on top of them, thus removing/killing my flowers. Besides Public Works and Homes/Buildings already in the way, is there any other full proof way of preventing new villagers from moving in on top of my garden? Thanks!

Not really.
I've lost so many bushes and flowers to move ins. You can try to force them away from areas by strategically placing PWPs, but that's mostly it.
Otherwise, I'd just say not to plant flowers around hazard areas in your town until you've gotten a good stock of potential replacements.
 
You can do a plot resetting trick by creating a new character (and not loading your mayor for the day). Run around the town and see if they plotted somewhere. If you like it, keep playing until you can save. Then you can delete that character whenever. If you don't like it, close the software and try again. The spot will be randomized as long as you haven't loaded an existing character (because it automatically saves when you do). Or you can do the PWP placement as stated above, or bury random items underground.

Oh, I didn't know you could do that actually. Thanks for the idea, I suppose I'll use this when deciding where I want the next villager placement! Thank you for the suggestion~.

@Yukari - Will do, I'll probably use PWPs as a safe haven for my flowers haha. As long as they keep the villager homes away!

The plot-resetting trick definitely works! What I do, and definitely recommend to do if you plan on keeping a town in perfect shape, is create a new, temporary character every day before I play to make sure a house plot isn't in an inconvenient place. Once you're done, you can just delete the character and carry on with your day. :)

I also do this every time I get a dream villager to get them into an optimal moving place. From my thousand-times-over experience with it, it doesn't pose any risk at all to your town (or save data, for that matter) and is super handy if you want to have a town go *partially* your way.
